Madekwe was raised by an ethnically diverse family. She has a mother who is English and her father is Nigerian. Her family was in the South London Council Estate and was a student at The Brit School aged 14. The Royal Academy of Dramatic Art is where she earned a BA degree in acting. While at school she appeared in a variety of stage productions such as King Henry V's Princess Katherine as well as Wuthering Heights.

Her credits in the theatre include Little Sweet Thing (by Roy Williams) as well as Flight Path (by David Watson).
Madekwe officially made her US debut with The Beautiful Life in 2009. The show aired just two episodes on The CW, and was later removed from the air.
